Principles and Practice of Analytical Chemistry

Preface to the Fifth Edition
It is twenty-five years since the first edition was published, and at the beginning of the twenty-first
century it seems appropriate to reflect on the directions in which analytical chemistry is developing.
The opening statements from the preface to the first edition are as relevant now as they were in 1975,
viz 'Analytical chemistry is a branch of chemistry which is both broad in scope and requires a specialised and disciplined approach. Its applications extend to all parts of an industrialised society.'
During this period, the main themes have remained constant, but differences in emphasis are readily
discerned. An increasing concern with the well-being of individuals and life in general has led to
initiatives for improvements in medicine and the world environment, and in these areas analytical
chemistry has particularly vital roles to play. The elucidation of the causes and effects of ill health or of environmental problems often depends heavily upon analytical measurements. The demand for
analytical data in relation to manufactured goods is increasing. For example, in the pharmaceutical
industry much effort is being concentrated on combinatorial chemistry where thousands of potential
drugs are being designed, synthesised and screened. This activity generates considerable analytical
requirements, particularly for automated chromatographic and spectrometric procedures, to deal with
very large numbers of samples. The determination of ultra-trace levels and the speciation of analytes
continues to provide challenges in the manufacture of highly pure materials and environmental
monitoring.
A recurring theme throughout the discipline is the sustained impact of computers on both
instrumentation and data handling where real-time processing within a Windows environment is
becoming the norm. Data reliability in the context of policy development and legal proceedings is also of increasing importance. Such developments, however, should not distract analytical chemists from the need for a sound understanding of the principles on which the techniques and methodologies are based, and these remain prominent features in the fifth edition. Whilst it would be true to say that no totally new analytical technique has gained prominence during the five years since the fourth edition, most of the established ones have undergone further improvements in both instrument design and methodology.

The introduction of high-performance liquid chromatography (HPLC)
to the analysis of peptides and proteins some 25 years ago revolutionized the biological sciences by enabling the rapid and sensitive analysis of peptide and protein structure through the exquisite speed, sensitivity, and resolution that can be easily obtained. Today, HPLC in its various modes has become the pivotal technique in the characterization of peptides and proteins and currently  plays a critical role in both our understanding of biological processes and in the development of peptide- and protein-based pharmaceuticals.
The number of applications of HPLC in peptide and protein purification
continues to expand at an extremely rapid rate. Solid-phase peptide synthesis and recombinant DNA techniques have allowed the production of large quantities of peptides and proteins that need to be highly purified. HPLC techniques are also used extensively in the isolation and characterization of novel proteins that will become increasingly important in the postgenomic age. The design of multidimensional purification schemes to achieve high levels of product purity further demonstrates the power of HPLC techniques not only in the characterization of cellular events, but also in the production of peptideand protein-based therapeutics. HPLC continues to be at the heart of the
analytical techniques with which scientists in both academia and in industry
must arm themselves to be able to fully characterize the identity, purity, and
potency of peptides and proteins.

Liquid-liquid extraction is the separation of one or more components of  a liquid solution by con­act with a second immiscible liquid called the solvent.
If the components in the original liquid solution distribute themselves differently
between the two liquid phases, separation will result.
This is the principle upon which separation by
liquid-liquid extraction is based, and there are a number of important applications of
this concept in industrial processes.

Liquid-Liquid Extraction Operations and Equipment

Liquid-liquid extraction is a process for separating components in
solution by their distribution between two immiscible liquid phases.
Such a process can also be simply referred to as liquid extraction or
solvent extraction; however, the latter term may be confusing
because it also applies to the leaching of a soluble substance from a
solid.
Since liquid-liquid extraction involves the transfer of mass from one
liquid phase into a second immiscible liquid phase, the process can be
carried out in many different ways. The simplest example involves the
transfer of one component from a binary mixture into a second immiscible
liquid phase. One example is liquid-liquid extraction of an impurity
from wastewater into an organic solvent. This is analogous to
stripping or absorption in which mass is transferred from one phase to
another. Transfer of the dissolved component (solute) may be
enhanced by the addition of “salting out” agents to the feed mixture or
by adding “complexing” agents to the extraction solvent. Or in some
cases a chemical reaction can be used to enhance the transfer, an
example being the use of an aqueous caustic solution to remove phenolics
from a hydrocarbon stream. A more sophisticated concept of
liquid-liquid fractionation can be used in a process to separate two
solutes completely. A primary extraction solvent is used to extract one
of the solutes from a mixture (similar to stripping in distillation), and a
wash solvent is used to scrub the extract free from the second solute
(similar to rectification in distillation).
